I am trying to lock the EXCEL Workbook to one user at a time and have the message pop up, "WORKBOOK is LOCKED by another user and is READ ONLY." Can someone one show me where I can change the settings to achieve this task.

Two options:
1. Store it in a SharePoint document library where require checkout is enabled.
Set up a library to require check out of files - Microsoft Support
2. Store it on a network share and do not use legacy sharing (Review | Share workbook)
