I am currently doing some research on Windows 365.  Can we connect a Windows 365 PC to Private Cloud so that users can access local resources, like a file server?

Yes only with the Windows 365 enterprise flavor, you can choose either AADJ or HAADJ method and choose Azure network to deploy your Cloud PC's into.
