I don't "KNOW", as I have never encountered this.
However, I might try a few things.
Microsoft released PC Manager

Install it and run all the trouble-shooting components.
When you downloaded DEFENDER did you ERASE the previous installer exe? Just wondering.
And, you have full ADMIN rights?
Checked the REGEDIT file and see DEFENDER properties?
Install another AV product, run a scan. Remove the "other" AV product. Download DEFENDER again.
Re-install DEFENDER. Thinking outside of the box and established BEST PRACTICES.
