Azure AD unblock users O365

What is happening is you are likely changing the cloud attribute. Since nothing has been changed on prem, the next time the sync happens from AADC, it is undoing that value.
The easiest way to do this would be to disable the user on your local AD. That would disable the users login to O365, but not delete anything for them in the cloud.

The problem is that I want that my users to be able to login local, but not on office 365.

If you are just selecting to block their access through the portal, it could be working, it just takes awhile to go through.
Typically Microsoft tells you it could take 24 hours for the setting to take effect.

The problem is that it just does not work even after 2 weeks.

If you move the user out of scope in syncing on AADC, it will remove their O365 account. Essentially it just unlicenses them.
That will delete everything after 30 days, but it would prevent access while everything was removed. You would just need to make sure you re-license the user to prevent deletion before then.
