By default Project uses working time to measure duration, so with the Standard calendar a "day" of duration is 8 hours and a "week" of duration is 5 days.
However, you can change this in a couple of ways. One is to enter duration in elapsed time. So for example a task with a duration of 2ed would equate to 2 calendar days or 48 hours. The other method is to create a custom Project calendar that has different working hours (e.g. 10 hour days or 7 normal working days).
As far as partial days appearing in the Duration field (e.g. 4.5d) it all depends on what time a task starts and finishes. This is easy to see by setting the Project options to display the time as well as the date. Note, this setting is not available if you are using Project for the Web.
Hope this helps.
