Access to Access
... and now, when I try to restart, having closed everything, I get this!
That appears to be a Windows error, which I don't recall seeing. It's not coming from Access, I don't think.
Make sure the accdb in the local folder has been made editable. There is a feature on files downloaded from the internet which marks them read-only. You can see that here, if it is.
I would recommend a reboot of the computer. When it restarts, check the accdb for that flag.
Keep at it; you'll figure this out.

Whatever is going on on that computer is not an Access specific problem and I don't know how to help with that. I toggle the 'read-only' to off, accept and close the properties dialogue box. I open it immediately and there it is again, read-only checked!

I also get this on starting the computer and occasionally at other times also. It doesn't seem to make any difference whether I say yes or no, it still returns randomly.

We're approaching the border between my knowledge of Access and the vagaries of Windows. It seems this is more a Windows problem than an Access problem.Please try one more thing.
Create a new folder on the C:\ drive, not in your users folder, directly on C:\
Put the accdb in that new folder, for example;
C:\TestFolder\youraccdbnamegoeshere.accdb
Make that TestFolder a Trusted Location.
Still no luck?
