I wouldn't be worried about the Proofpoint if you're making any changes on mail flow.
if you're moving out of SOPHOS Endpoint Adv deploying M365 Defender just stick with MSFT recommendations https://learn.microsoft.com/en-us/microsoft-365/security/defender-endpoint/migration-guides?view=o365-worldwide and https://learn.microsoft.com/en-us/microsoft-365/security/defender-endpoint/switch-to-mde-overview?view=o365-worldwide ,
