Thank you Paul. If we wanted to use it as an on premises relay though would we need to configure the hybrid feature?

Nope. You will need to configure the on prem Exchange server to allow the incoming connection and then you can either send it straight out via MX lookup or you can relay it through O365 using this article: https://support.office.com/en-us/article/How-to-set-up-a-multifunction-device-or-application-to-send-email-using-Office-365-69f58e99-c550-4274-ad18-c805d654b4c4
