Is it possible to create a DLP custom policy based on file extension detections. For example: *.exe, *.txt ?

Yes, You can create custom policy DLP policy for supported file extensions.
Follow this procedure to create one.
Data loss prevention>Create policy> Custom policy> (Fill the necessary details ) and jump to> policy setting: (create or customize Advance DLP rules)>Customize Advance DLP rules >create rule> Conditions> add Conditions(File extension is).
