What is going on in the background when you add an SMTP Proxy address to a user account?

When updating a proxyadress of a e2k3 user mbx from an adu&c set on a admin workstation, we had latency going up to ~40sec. When applying the regkey "preferred sa server" with e2k3 sp2, the latency was 5 sec. Good news ! BUT for some clients, Latency can take 5sec for the first update and 5 mn for the next updates or the same or different user. Do you Have any ideas ? or have some advices to investigate further ?
