ceantuco /PrepareDomain is required to run if the objectVersion(Default) has been raised. It is then necessary to run /PrepareDomain for each domain which contains Exchange-related objects (or to use /PrepareAllDomains to prepare all available domains).
The version has been raised between Exchange Server 2019 CU10 (13241) and CU12 (13243) and therefore, /PrepareDomain was enforced to run. It remains the same with CU13 (13243) and I don't expect that a /PrepareDomain run is enforced. However, it could be enforced if Exchange setup isn't able to read the current version from AD.
