ADPrep from 2012R2 to 2019

I was running all 2012R2 domain controllers and installing a new 2019. ADPrep is failing to find CN=Keys,DC=Domain,DC=com. I am wondering do I have to run adprep from 2016 before I can run 2019?

No, you don't have to.

This will all be sorted out automatically once a Windows Server 2016 or above domain controller takes over the PDC FSMO role.
