Are there key differences, challenges or best practices when doing Live Events via the three available platforms?

Yes, depending on the level of production you'd like. Teams is great for a quick event (without an external encoder) and includes moderated Q&A. Stream is the right choice if you don't need conversations and/or want to embed it in your SharePoint site. Yammer is the right move for richer conversations, broadcasting the event across your organization, and the video to live within a Yammer group before, during, and after.
