We have an MS Exchange 2003 SP2 Server (native) and we have windows XP SP1 & SP2 Clients, if we use the Exchange DST update tool, do we have to do anything to the client pc's besides the XP hotfix

You must apply the 931836 OS update to the XP SP2 machines. Please review KB 914387. Windows XP SP1 is not in support any more.
